AnotationTraditional two-dimensional cell cultures as model systems have significantly contributed to cancer research but are very distant to the real tissue and tumor environment, particularly in terms of cell interactions, tissue factors and genetic heterogeneity. To overcome these limitations, organoid systems have been developed to better model real architecture and complexity of tissues. The Blood Cancer Research Group at FNO and LF OSU has been successful in studying multiple myeloma (MM), which is characterized by the presence of numerous aberrant plasma cells in the bone marrow (BM). Importantly, the BM microenvironment has a very significant effect on the pathogenesis of MM and resistence to therapies. Therefore, the aim of this project is the development and application of organoid cell systems for the modeling of BM microenvironment in the development, progression and drug resistance of MM. The introduction of a unique organoid technique that mimics the BM microenvironment in laboratory conditions will mean not only a considerable shift in methodology, but will also allow for testing a number of hypotheses both in the field of basic research and the analysis of new therapeutic approaches that can be difficult to validate in classical cell cultures.
