AnotationThis project will focus on the development of an analytical methods for the determination of serum concentrations of selected drugs used in the treatment of sclerosis multiplex. The development of the method will be performed by liquid chromatography coupled with tandem mass detection. Great attention will be paid to methods of biological matrix treatment in order to achieve fast and simple extraction procedure with the highest possible extraction yields. During the development of the method, the conditions of chromatographic separation will be tested as follows: the composition and flow rate of the mobile phase, the type and temperature of the chromatographic column and the injection volume of the sample. Furthermore, the parameters of the mass spectrometer will be tested: temperatures and gas flows, setting of ion optics and finding identification and quantification MRM transitions to obtain low values of detection limits (LOD) and quantification limits (LOQ). The developed method will then be validated according to the rules of the Food and Drug Administration (FDA) for the validation of bioanalytical methods as follows: matrix effect, linearity, working range of the method, limits of detection and quantification, accuracy, precision, interference and stability.
